Research
Structural characterization and functional analysis of an anti-cocaine monoclonal antibody for treatment of cocaine addiction
My laboratory is currently collaborating on a therapeutic anti-cocaine antibody project with the laboratory of Dr. Andrew Norman. We have characterized the post-translational modifications and analyzed the heterogeneity of this monoclonal antibody, as well as developed methods for generation and purification of antibody fragments, including the therapeutically relevant Fab fragment. We also developed fluorescence quenching methods to measure binding affinities of the antibody and its fragments for cocaine and its metabolites and derivatives. We are currently characterizing the stability of the intact antibody, crystalizing the Fab fragment, and determining the protein and glycan heterogeneities and post-translational modifications of this monoclonal antibody to seek FDA approval for human clinical trials.
Another long-standing goal of the laboratory has been the determination of the structures and functions of the cell membrane and soluble ecto-nucleotidases. One family of these enzymes are collectively called ecto-Nucleoside Triphosphate Diphosphohydrolases (eNTPDases), and another is a structurally unrelated nucleotidase is called SCAN (Soluble Ca-Activated Nucleotidase). The ecto-nucleotidases are important for the regulation of many physiological and pathological signaling processes under purinergic signaling control, including pain perception and maintenance of hemostasis via hydrolysis of the platelet activator, ADP.
Related to the nucleotidases, I also have ongoing collaborations with Dr. Peter Roach (Indiana University) using the SCAN nucleotidase to characterize glycogen phosphorylation, and with Dr. Neil Ayres (Department of Chemistry, here at UC), testing the suitability of coatings his laboratory synthesizes for making materials biocompatible with blood.
