Manoj K Pandey , BS,MS,PhD

Dr. Manoj Pandey's laboratory is working in area of IgG and C5a-mediated diseases including recurrent spontaneous abortion, autoimmune, allergic, and Gaucher diseases. Major findings of his research include 1). Identification of a critical role for IgG3 in mixed lymphocyte reaction blocking antibodies (MLR-Bf), which are critical for maintenance of human pregnancy. 2). Running a double-blind randomized trial to evaluate the efficacy of paternal T and B cell immunotherapy for development of MLR-Bf and its relation to the success of pregnancy in women with RSA. 3). Identification of the link between FcgRs and measles-specific IgG, which allows for augmented entry of measles virus in MOs and Mɸs so that the virus can escape from anti-host immune responses 4). Demonstration of a novel anti-inflammatory pathway in which IgG1 ligation with FcgRIIB and dectin-1 blocks C5a mediated inflammation in a mouse model of autoimmune disease. 5). Identification of staphylococcal complement modulators (SCIN-B, SCIN-C, Efb and Efb-like), which abrogate C5a-mediated inflammation. 6). Using adoptive transfer and in vitro culture of CD4+CD69+ T cells with pulmonary myeloid DCs (mDCs) he defined the dual role for C5a in allergic asthma, one which suppressed Th2 immune responses during allergen sensitization at the DC/T cell interface and the other, which enhanced airway inflammation in an established inflammatory environment. 7.) Dr. Pandey has recently shown that not only Mɸs, but also DCs and CD4+T cells of Gba1 mouse model of Gaucher disease (9V/null) suffer from excess GC storage and express increased levels of MHCII and co stimulatory molecules. These cells further show increased production of pro-inflammatory cytokines and chemokines, which is associated with increased tissue recruitment of Mɸs, DCs, and T cells. These findings are novel, as immunological alterations described in Gaucher disease focus on Mɸs. The nature and the possible role played by other immune cells have not been explored. We have performed a detailed phenotypical and functional analysis of the main APC subsets, including Mɸs, DCs, and T cells, suggesting that additional cell populations, i.e., DCs and CD4+T cells are critical effector cells driving inflammatory responses that lead to Gaucher disease manifestation.

Using mouse model and human patients with Gaucher disease, the major focus of investigation in the Dr. Pandey laboratory is to study the 1) Role of immunological cells i.e.,dendritic cells, macrophages, neutrophils, T,  and B lymphocytes and their competence for glucosylceramide (GC) accumulation. 2) Using these cells to evaluate the impact of GC in induction of several innate and adaptive immune responses. 3) Uncovering downstream signaling critical for immunological inflammation in Gaucher and other lysosomal storage diseases.
